Retroviruses: Poorly Understood Agents of Change

Chronic inflammatory diseases have been skyrocketing in incidence in the past quarter century.

The details explaining how retroviruses in today�s biological therapeutics including

vaccines are contributing to autoimmune, neuroimmune disease and cancer are complex.

Although I�ve spent my adult lifetime studying how retroviruses contribute to these diseases,

paring down the complexities into basics is a daunting task.

In our book, Plague, Kent Heckenlively and I detailed the science and cover up surrounding

my team�s 2009 discovery of a new family of human retroviruses related to mouse leukemia

viruses, associated not only with cancer but with Autism Spectrum Disorders and Chronic

Fatigue Syndrome.

In Plague, my coauthor and I detail the science behind the discovery.

Scientific research is not simply a study set in a defined space or time, but a lifetime

of detailed observation and learning�a lifetime of forming hypotheses and modifying those

hypotheses as technology and learning inform new discovery.

Science is never settled as we learn each day and discover things that were once considered

impossible.

However, science in the 21st century is more complex than ever in human history.

Kent Heckenlively is a sixth-grade science teacher.

In order to tell my story in a way everyone could understand, who better than a sixth-grade

science teacher to help explain the intricacies?

Or so we thought.

The reviews of Plague include one from a doctor who said the science was �too complex.�

As this is such a critical topic in human heath, I want to make it as simple as possible

so that everyone can understand.

What are retroviruses?

Retroviruses are classified in a group of RNA viruses called RNA tumor viruses.

They are called �retro� because they only have an RNA genome and function differently

than other viruses.

In most viruses, DNA is transcribed (or written) into RNA, then RNA is translated into protein.

Retroviruses, on the other hand, work differently.

A retrovirus works by reverse transcribing, that is �writing backwards� into DNA by

using an enzyme only retroviruses encode called, �Reverse Transcriptase� (RT).

The DNA form of the virus is called a provirus.

The provirus is then inserted into the DNA of the host using another enzyme encoded exclusively

by retroviruses called �Integrase�.

(IN).

Integrase cuts open the DNA and then pastes the provirus into the cellular DNA where the

provirus lives for the life of the cell.

In addition to RT and IN, retroviruses encode a few other key genes important to make a

virus particle called a virion.

The envelope gene called env and gag encode the proteins that form an envelope and capsid,

which surrounds the RNA genome.

The RNA genomes of retroviruses are between seven and twelve thousand bases (7-12 kilobases,

kb).

The human genome contains approximately three billion base pairs.

(RNA is single stranded, while DNA is double stranded, hence �base pairs.�)

A retrovirus virion is approximately 100 nanometers (nM) in size and can only be seen by an electron

microscope.

Importantly, the provirus cannot be made into an infectious viral particle without using

the machinery of a dividing cell.

This is illustrated in the dark parts of the membrane of the cell where the virus is budding

out of the cell taking the lipids from the cell membrane to complete the virion.

Here, there, and everywhere

Essentially, all animals have retroviruses integrated in their genomes.

Birds, monkeys, cows, pigs, cats, dogs, mice and fish all have retroviruses encoded in

their genomes; even plants have retroviruses.

Vertebrate genomes harbor thousands of endogenous retrovirus (ERV) elements that display a structure

close to that of the integrated proviral form of exogenous retroviruses (gag-, pol-, and

env-related regions flanked by 2 LTRs) but the genes are mutated so that they are thought

not to be able to produce and release infectious particles.

That is, ERVs most likely are the remnants of past infections of the germline by ancestral

retroviruses which have been crippled by the immune system of the host.

This means that the retroviral genes are defective and no longer release infectious particles.

As much as 15% of the human genome is made up of ERV human retroviruses.

In animals, exogenous retroviruses are responsible for some of the deadliest diseases known.

Yet, it wasn�t until 1980 when Poiesz and Ruscetti isolated the first human disease-causing

retrovirus, then called Human T-cell Leukemia Virus as it was shown to cause an aggressive

cancer called Adult T-cell leukemia (ATL).

In fact, when my mentor and colleague of 35 years, Frank Ruscetti, joined the National

Cancer Institute (NCI) in 1975 to study human disease causing exogenous retroviruses, he

was told by NCI scientist John M. Coffin not to bother as they did not exist.

Although retroviruses have been an important part of human evolution as the placenta evolved

from ancestral retroviral envelope genes 25-40 million years ago, envelope genes from both

exogenous and endogenous retroviruses, aberrantly expressed in humans, have been shown to be

responsible for the development of many chronic diseases.

The incidence rates of these diseases are skyrocketing in 21st century America and include

prostate cancer, breast cancer, leukemia lymphoma, multiple sclerosis, and amyotropic lateral

sclerosis (Lou Gherig�s disease).

Expression and mode of development

Many factors are important in the development of diseases associated with retroviruses.

The expression and mode of transmission are keys to disease development.

We have learned a great deal about the types of diseases from 40 years of study of the

mechanisms of disease development from animal and human retroviruses.

In mitotic transmission, the provirus is dormant or defective and the integrated proviral form

of exogenous retroviruses (gag-, pol-, and env-related regions flanked by 2 LTRs) are

not expressed.

In this case only the daughter cells carry the retroviral genes and if not expressed

these endogenous or exogenous retroviral genes remain dormant for years and do not usually

contribute to disease until much later in life as the immune system weakens.

During infectious transmission, the complete virion is produced with many thousands of

virions infecting many neighboring cells and spreading from person to person�both cell

free and cell associated�via blood and body fluids.

Infectious transmission of HIV drove the AIDS epidemic of the 80s and 90s including transmission

from infected cells in a contaminated blood supply and the activation of dormant retroviruses

by heavy metals, co-infections and inappropriate vaccination of HIV infected individuals.

Xenograft approaches commonly used since the 1950s in studies of human cancer, autoimmune,

and neuroimmune disease promote the evolution of novel retroviruses with pathogenic properties.

We now appreciate that it is the use of xenograft technologies in the development of vaccines

and biological drugs and genetically modified organisms (GMOs) that have accelerated the

spread of animal retroviruses into humans, a process known as zoonosis, whereby an animal

retrovirus jumps species, learning to evade immune mechanisms

of humans and thereby causing disease.

it's a 20-second clip I would have played it before this video so the

trailer is basically to show the audience some context clear up some

scenes from the other trailer but remember the trainers are misleading

until the actual episodes have air the idea of a trailer is to show a highlight

reel or the best parts and hype you up for the episodes so you actually watch

it so let's get into this breakdown so the trailer begins with a scene of a

shock traveling on the road the camera pans over and we see the flash is

lightning for the first time the flash has a collision with the truck and

pursue min it's the flash then we see the person with a bunch of apples around

his feet we see the Speed Force pulsing around him so we can presume there is

Barry Allen but on the record I'm gonna say 99% I think it's Barry Allen we see

Cecile for the first time in a while telling Joe Iris Wally West and Cisco

you need to prepare yourself so bazoom Barry is going to be lost so iris sees

her fiance for the first time since the season finale where he told her to keep

running and never stopped running where he sacrificed himself and went into the

Speed Force to save central city this would be such an emotional episode for

the couple in many ways iris West says to him Barry

you're home which it's going to be a cool scene this makes sense because in

the first scene Barry Allen and being naked the people in the truck would have

most likely call the police hence why he ends up at City PD and cecile Joe West

girlfriend has him instead of going to iris West because he doesn't know who he

is the only thing here is how will the show - explain where Barry Allen's been

for six months and how he turned up in the middle of nowhere naked scene as

he's supposed to be the CSI for the CCPD so Barry looks broken we know this

already from an interview I think it was during Comic Con when Grant Gustin where

he explained that we're not really going to see much of Barry Allen inside the

Speed Force we're just going to see ramifications act throughout the whole

season and it also went on to say that Barry Allen would have lived his life

infinity times because time inside the Speed Force is linear so it does make

sense that Barry Allen would be broken I mean I'm taking it from the clip that he

doesn't even know who he is although I did think they would play on

iris West being his lightning rod and that the fact that they will always find

each other the next scene that we see is the samurai that says bring me the flash

the samurai takes iris west up into the air so they are respecting the comic

book mythology or see the flying samurai obviously they're called samurai in the

comic books and obviously there's multiple amounts of these but obviously

this is a TV show unlimited budget so it's pretty cool that it's kind of the

drama is focused around iris West again seeing as she is the most

important person to Barry Allen and that is most likely what would spark him to

remember who iris is to him Barry gets some tests on him most likely it would

have been at STAR Labs because if they took him to a normal Hospital it would

have been a bit weird and they might have found out who is so we see that

Barry does have a beard in the scene which is pretty interesting I'm glad

they kept this a secret obviously these scenes would have been

done inside just TV studios rather been done outside in the open and then you

get obviously the beat behind the scenes people ruin it so Cisco says I made you

a suit proven that he didn't manifest it within the Speed Force which is kind of

interesting I know people will most likely not like this because they would

have afford Barry to come out the Speed Force and manifest the Speed Force the

new suit the Speed Force but we did see in the first scene that whoever this is

it most likely will Barry will be naked so it's pretty cool that we've got a

clarification of where the suit came from and Cisco's like want to take it

for a spin the best scene in the trailer is when Barry starts writing lots of

equations on the board we even see light in his eyes it reminds me of sabotage

and future flash kind of because his hair looks long but what's interesting

is in this scene he doesn't have a beard but it's supposed to be from the first

episode so like I said guys the trailers are misleading we don't even know if

this scenes actually going to be from one because these are teased scenes and

it's misleading it could even be some scene from the speed force me personally

I would be shot if we don't get at least one scene from his time inside the Speed

Force Barry smashes through the prison within STAR Labs which causes an

explosion we can see this in the trailer and we see that Joe is on the floor now

this is very interesting because this shows us either one of two things how

powerful Barry Allen and the flash has got or how fast he's potentially going

to be because remember guys ebar thorne was in themselves multiple times and he

never attempted to get out once we've seen many of people attempt to get out

of themselves in other seasons when Barry Allen's been locked up in there he

couldn't get out even after trying so this does give us a cheeky wink from

behind the camera that Barry Allen is going to be getting a lot stronger which

is going to be interesting because let's go back to the actual burial and writing

them equations on the wall so obviously he's in some kind of Speed Force trance

he's gonna have like a lot more knowledge because he would have lived

his life multiple times has said drawing up that interview so he's gonna be a lot

smarter and wiser he just needs someone like Cisco to decipher these equations

because these equations are not currently shown for no reason these

equations are going to be something he would have seen the future he would have

seen that everything he's going to have in Affinity brain power but he won't be

able to use it because of these obviously being stuck in the Speed Force

stuck in some kind of trance and a pretty cool scene is where Cisco says

the flash is back and you notice behind him that the current flash shoot is in

the background so at this point wally west hasn't put on the suit and hasn't

pretended to be the flash and it hasn't been ripped up like we've seen in behind

the scenes footage so it's quite interesting interesting because like i

said guys this is a 30-second trailer so we don't know what part is what what

part is what part the trailer and we don't even know if this is actually from

episode one it could even be a glimpse of episode two Barry is running out of

control he can't handle his speed as we bouncing around STAR Labs like a nutter

and we also see smashes outside of the prison which is kind of interesting so

I'm really interested to see how much faster Barry Allen will actually be when

he gets out of the speed not sorry out of the kind of prison that they put him

in to crime to contain him as I think Barry doesn't know who he is

but what's really interesting is right at the end we got a Wonder Woman like

song right after the credit not credit right after the trailer ends we get the

new flash logo it's all pure red and we get the Wonder Woman type music let me

know guys in the common box down below do you think it's actually a one-to-one

music for me it sounds slightly like music played during the Wonder one I

think it's fantastic that we're getting a new flash logo which is pretty cool as

it is called the flash reborn which is basically a nice egg comic-book

reference to the flash rebirth in the comics so it would make sense that they

changed the flash logo and make it actually read for once so I'm really

looking forward so the flash season for after this trailer this trailer really

does give us an insight into what we can expect for the flash season four because

let's be honest guys before this trailer everything was stale do you notice that

we didn't get any caitlin snow in this trailer and let me know guys in the

comment box low did we get Caitlin so in a trailer

I believe there's only one clip of Caitlin where she just looks her normal

self she doesn't look like the actual clip where we actually seen in the

comic-con trailer and it just kind of makes them match parts from all the

trails and we have got clarification of some scenes now let me know guys in the

comments below what is your favorite scene from this I really did like the

opening scene where he collided with the truck and he was naked and the fact that

he's not coming out knowing who he is so it's gonna have

context he's actually going to have ramifications from being inside the

Speed Force because if we go back to season two in the Kevin Smith episode

where he was in the Speed Force he spoke to his mom he realized everything that

that running away with the dinosaurs episode he there

really any ramifications of being inside the speedforce and also when Kid Flash

was in the Speed Force because Savitt are tricked him he was in the Speed

Force and there was no ramifications though except for when he was in there

it looks like we're finally going to get to see the ramifications of what being

inside the Speed Force actually does to a person and Barry Allen is going to

come out such a different kind of person he's going to be wiser smarter more

intelligent apparently funnier more like a flash season 1 in terms of joking and

cracking jokes and being wiser but he's also going to be more mature because

they need to give him some character development throughout like the flash

season 3 Barry Allen didn't really get much character development

except for realizing that everything happened because he created flashpoint

so I'm really looking forward to seeing the ramifications of being inside the

Speed Force but I really hope we actually do get some Speed Force

ramifications that play out throughout the whole season and don't just fizzle

out from the first couple of episodes anyway guys this has been a review from

10 Best Rotator Cuff Pain Stretches - Duration: 10:55.

hey everybody it's doctor Jo and Bear, and today we're going to show you the

top ten stretches for the rotator cuff. so let's get started.

so these top ten stretches are my favorite top ten. there's no research

behind it it's just what I found has worked best for me and for my patients.

so by the term rotator cuff, that usually means four main muscles. so

supraspinatus, infraspinatus, teres minor and subscapularis, but when you really

think about the rotator cuff, the rotator cuff has a lot to do with the scapula

and the shoulder blade, and there's like 20 muscles that attach to the scapula, so

when we're doing stretches, we're not just stretching those rotator cuff

muscles, we're stretching all the muscles around that scapula and shoulder blade

because if one of those are out of balance, it's going to make everything

else out of balance and cause a lot of problems with the rotator cuff. and just

a reminder for the stretches when you're doing a stretch, you want to hold it for

at least 30 seconds and do it three times, so that really lets those muscle

fibers go, help stretch them out that Golgi tendon organ, or the GTO, you really

need that 30-second hold for it to relax and let go, so 30 seconds three times. so

the first stretch that we're gonna do is pendulums. pendulums are one of my

favorites for the shoulder. what it does is it really just helps open up that

joint, loosen up those muscles, really kind of get them warmed up for you to

stretch. so let's take a look at that. for shoulder pendulums, you're just gonna

dangle your arm down towards the floor, you're not actively gonna move your arm,

but you're going to use your body to move your arm making circles and then

reverse it the other way.

for shoulder arm swings front to back, just let your arm dangle down, you're not

actually moving the arm but you're moving your body to make your arm move.

for shoulder arm swings from side to side, just dangle your arm down. keep it nice

and relaxed, you're not actually moving your arm but you're moving your body

side to side.

the next stretch is just going to be a scaption stretch, so scaption is

basically not flexion out in front of you, not abduction out to the side, but

it's kind of right in the middle, and just doing some scaption stretches going

straight up and down is a really great way just to kind of again stretch out

that shoulder, but not holding the stretch just kind of getting those

muscles warmed up and moving, so let's check those out. for shoulder scaption

active range of motion, put your thumb in an upward position and keep your arms

straight, it's not completely out to the front and it's not out to the side, it's

about in the middle at a 45 degree angle, and just bring it up to about 90 degrees.

number three. so stretching with a stick or a PVC pipe, you can use you a cane if

you have a cane, is a great way to really get that extra stretch especially maybe

you're not able to quite get that movement yet, so we're going to do a

flexion stretch with a stick, so let's check that out.

for shoulder flexion passive range of motion with a stick, you can use a cane,

or a broomstick, or PVC pipe if you have some. your arm is just going to go along

for the ride, so the other arm is going to do all the movement it's just going

up and down, and then the other ones just sitting on top.

number four, now we're going to go into an abduction stretch with a stick, so

again I really like using the stick it kind of helps get into that stretch if

you're still in a passive stage, or an active assistive stage where you can't

really move it on your own yet, like you're not supposed to because of a

precaution or just because you can't because it still hurts a lot. using the

stick really helps move that shoulder and that arm for you without you having to

actively doing it. so let's check out. for abduction for shoulder abduction passive

range of motion with the stick, you can use a cane, a broomstick, or PVC pipe if

you have some. your arm is just going to go along for the ride and the other one's

doing all the movement. so it's pushing the arm up, just set your hand on top of

the cane or the PVC pipe with your thumb pointed upwards, and then just push your

arm up as comfortably as you can.

number five is going to be external rotation stretch with the stick. so the

same concept you're using that stick to push through that motion to really get

that stretch and a lot of times again it's too painful to do or maybe you're

not supposed to actively use those muscles yet if you've had a surgery or

something, so this is a great way to get that stretch and that passive or that

active assisted range of motion. so let's take a look at that. for shoulder

external rotation passive range of motion with a stick, you can use a cane, a

broomstick, or PVC pipe if you have some. the arm is just going to go along for

the ride, and the other one's going to do all the movement. bend your elbow and put

it by your side, keep it there the whole time, try not and let it come out, then

hold on to the pipe with your thumb up and then push outwards and come back in.

so remember make sure that elbow is not coming away from your side.

number six wall slides. I really love wall slides because again you're using

something to kind of help support your arm, so it's just like it sounds, you're

sliding up the wall, but it's great because instead of actively doing it all

by yourself, you have the wall to kind of help support your arm, and you also have

that wall to kind of lean against and push up against to get that extra

stretch, so it's a great way to get that stretch. let's check it out. for a

shoulder wall slide, place your hand on the wall with your thumb facing outwards,

slide your hand up the wall as far as you comfortably can.

number seven internal rotation stretch with the towel. this one's kind of tough.

it's a little bit uncomfortable, but it's going to be the one that helps get that

arm up behind your back, that internal rotation motion. it's not always

comfortable sometimes a little bit painful if you've had that rotator cuff

injury, this is usually the hardest one to get back, but using a towel really

helps stretch and get that movement back. so let's take a look at that. for an

internal rotation towel stretch, roll up a towel and with the hand that you're

not going to stretch with, put it up and behind you. then the one you want to

stretch it's going to go down behind you and then you're going to pull upwards. so

I'm gonna turn around so you can see it.

number eight. another great way to get that internal rotation, but not maybe

going back behind you but just getting that internal rotation stretch is what

we call a sleeper stretch. this one's really great too, and you can use your

other arm to kind of help push through it which is nice again it's a little

uncomfortable, but you know you don't want to be in pain while you're doing it,

you just want to feel a lot of tension while you're doing it.

so let's check it out. for shoulder internal rotation sleeper stretch, the

arm that you're going to stretch is on the ground. put your shoulder and elbow

at about a ninety degree angle, and then take your other hand and push it

downwards towards the floor until you feel a stretch.

number nine a corner stretch. this is really just to help open up those chest

muscles .it's a great way again to get that stretch using the wall to kind of

help push through that stretch and all these muscles in here. again some of them

might not specifically be those rotator cuff muscles, but they're muscles that are

really important to keep stretched and loose and help that whole rotator cuff

in general, so let's check out the corner stretch. for a corner chest stretch, place

a hand on each side of the corner and one foot in the corner then push in and

hold the stretch.

number ten a prayer stretch, or in yoga sometimes they call it the Child's Pose.

this is another one of my favorites because if you can get comfortably in

this position you can just relax your whole body and let your shoulders get

that stretch. you can go a little bit side to side or you can just go straight

down for that shoulder flexion. I really really like this one so let's check it

out. for the prayer stretch, start off sitting on your feet and then you're

going to push your arms straight out in front of you on the ground holding the

stretch.

so there you have, it those were the top ten stretches for the rotator cuff. if

Top 5 Wednesday - Books to Read Without a Synopsis [CC] | The Book Life - Duration: 4:25.

Hey guys it's Sarah Jane,welcome back to my channel. Today I am here to do a

Top 5 Wednesday. Top 5 Wednesday is run by Sam from Thoughts on Tomes I will

leave a link below to Sam's channel and also to the Goodreads group. Before we

get into it I just need to tell you guys something super quick, if you're not

interested just fast forward but basically I told you guys on Twitter

that I would be doing top 5 Wednesday so that's why I'm here filming at stupid

o'clock, basically I did my face I didn't do my hair as you can tell and went

outside and I found a really sick hedgehog and I ended up taking him to

the Wildlife Hospital where they're gonna hopefully make him better and I've named

him Hagrid, so fingers crossed he gets better but basically I wasn't expecting

to do top 5 Wednesday today after that but I told you guys I was gonna do it so

damn it I'm gonna do it. Even if my face looks like a moon

because that's how dedicated I am. So this week's topic is books you should

read without a synopsis, that is fairly self-explanatory it's books that you

should go into pretty much blind. So my list of books that you should read

without synopsis are not going to be in any particular order but let's get

started with number 5. The first book that I want to talk to you guys about is

the Graces by Laure Eve. I think this is a good book to go into blind because I

typically didn't know a lot about this book when I picked it up, all I knew is

that everybody in town is obsessed with the Grace's. It's a book that's got a bit

of a witchy vibe but it's not heavy heavy on the witchy it's a bit like

Gossip Girl meets charmed / the craft that's all you need to know, pick it up.

The next book on my list is The Big Lie by Julie Mayhew. The main reason that

I think you should go into this book without reading the synopsis is because

the synopsis may put you off if you're not typically a reader who reads this

sort of book. Because I can't sell you this book with a synopsis I'm gonna ask

you one question and that's gonna be what would the world be like for a

teenage girl in today's society if the Nazis had won the war? that's all you

need to know. The next book on my list is Rachel Vincent's Menagerie. Now this one

is a tricky one for me because actually I think the synopsis of this book is

incredibly strong it's what sold the book to me, but I feel

like if you like dark fantasy and you like fantasy that's set in a

contemporary world you'll like this. Fantasy creatures, dark mythology

a harsh harsh world - sign me up. The next book on my list is one that I

think you guys will potentially find a little bit amusing and that is The Raven

Boys by Maggie Stiefvater, now you have potentially heard a lot about this book

maybe you have read the synopsis maybe you haven't, I feel like you can read

this book without reading the synopsis simply because I'm now three books into

the Raven cycle and I still don't really know what's going on. And even though

when I read the back of this book and I read the synopsis it does give you a

hint of what goes on with the main character there's so much other stuff

that goes on I feel like this is a book that you can pick up if you just simply

like YA, you like books that get a little bit creepy and you like

character-driven stories. I honestly don't think reading the synopsis will

help you so you may as well just read it anyway. And the final book that I

recommend picking up without a synopsis is Maybe Someday by Colleen Hoover.

Maybe Someday by Colleen Hoover is a comfort food book it's a book that you

can fly through, its a book that gives you all the feels and it's a book that

doesn't have problematic content. Disclaimer - in my opinion it doesn't have

problematic content in comparison to some of her other books but it may do

that I've forgotten about and if it does I'm sorry. Some of Colleen Hoover's

other books are a bit problematic some of them are very problematic

and for that reason I'm not a huge huge lover of her books like I used to be but

this is one that's just it's just lovely. It's a love story that still has some

angst it still has some heartbreak but it's lovely and it's just it's just

comfort food it's comfort food reading when you need a book like that I

recommend picking this book up. It's a contemporary, it's a love story and

sometimes I feel that's all you need to know. So thats it for my top 5 Wednesday,
